Marvel will be releasing two Avengers movies over the next two years, Infinity War and an unnamed sequel. The mad titan Thanos will most likely assemble the "Infinity Gauntlet" by collecting all of the individual stones, giving him God-like powers. In what order will he do it? Some of this has been spoiled by the previews. In one of the most recent trailers, we see him entering Wakanda with at least 4 stones, possibly 5 (we can't see what's on his thumb). This spoils significant information about the plot. We know what the first four will be, but not the exact order. We know what the last two will be, but not the order. There is also the small probability that he never actually completes the set.
We have long debated what would be the optimal order to assemble the gems. At first it seemed like the choice should be to assemble them in order from easiest to hardest to acquire. Then each time the degree of difficulty goes up, he'll have more powers to aide in his success. The problem is, we don't know how the gems affect each other. There may be a specific order in which they need to be acquired. The reality stone might be the easiest get, but the power stone is the most important.
1. Power Stone: It becomes clear in Guardians of the Galaxy that Thanos is very focused on getting the power stone. That would seem to be his first priority, and it makes some sense why he needs that one before going after the rest. It would make fighting the Asgardians easier, and certainly a stone he'd like to get before facing the Avengers. Thanos will begin his gauntlet quest in Xandar, where he will get the most powerful of the stones. The Guardians will be there helping but will fail. Thanos will leave Xandar and the Guardians will be in pursuit.
2. Reality Stone: The reality stone is currently held by the Collector, and should theoretically be the easiest stone for Thanos to acquire. It's likely that it has been CGI'd out of some of the trailer footage, with a new trailer showing at least 4 gems on the gauntlet when he arrives on Earth. I've become reasonably certain about the order that he'll be acquiring the stones, but my greatest uncertainty is the reality gem. He may go visit the Collector before or after confronting the Asgardian ship, it could even be before Xandar.
3. Space Stone: Loki has the tesseract and will give it to Thanos. That will happen. We even see him handing it to Thanos in one of the trailers. The bigger question is, will Thanos kill Loki after this exchange? I'm sure the big guy isn't happy about how that New York thing played out and how Loki did not return the mind stone to him. Also, it would make travelling to other locations for the other gems easier. What will most likely happen is that Thanos will fight Iron Man, Spider man, Doctor Strange, and the Guardians of the Galaxy on Titan. After a flawless victory he will head to Earth.
4. Soul Stone: It's the one gem that Marvel has not officially confirmed to us it's location, but if Heimdall is the one who has it, then we have received plenty of clues. And if Heimdall is the one who has it, that means both the space and soul stones are currently together on the Asgard refugee ship. We are pretty sure that his last stop before coming to Earth will be his home planet of Titan. Either he will go home after destroying the Asgard ship, or the ship will already be close to the planet. We believe there's a strong probability that Heimdall will save the souls of his fellow Asgardians inside the soul stone before the ship is destroyed and Thanos gets the gem. ARRIVE ON EARTH
We know there will be a giant circle ship hovering over New York, probably right over Dr Strange's home and we know there will be a battle in Wakanda, probably in that order. This would suggest that he's going to get the time stone, then the mind stone. However, we also believe that Strange is going to hide the mind stone. Thanos will arrive in Earth with 4 stones.
5. Mind Stone: In the first Avengers movie, Thanos loaned the mind stone to Loki to help him invade Earth. As a result, Thanos no longer has it, Vision does. If your goal is to assemble them all, why loan any of them out to untrustworthy people? Perhaps the mind stone is the least important to the Gauntlet. There will probably be an attempt to remove the mind stone from Vision's head before Thanos arrives, but it may not work. If the stone can't be removed from his head without killing him, then the safest place for him to be is Wakanda. The stone will be removed and captured in Wakanda one way or another.
6. Time Stone: There is a scene in the trailers where Dr Strange is being tortured by Ebony Maw from the Black Order, probably because Strange hid it somewhere (like in another dimension) and they need to extract the location. The good doctor may crack under torture, he may not. Thanos could get the time gem in New York, or maybe never at all. There is a non-zero chance that he never completes the set of infinity stones, and the time gem might be the only one which could be used on its own to defeat Thanos in Avengers 4.
